摘要
目的筛选鲨鱼再生肝组织的差异表达基因。方法通过条纹斑竹鲨鱼肝脏2/3部分切除模型建立获得正常及再生24 h肝组织;采用20条10碱基随机引物和2条锚定引物通过差别显示逆转录-聚合酶链反应(differential display reverse transcription polymerase chain reaction,DDRT-PCR)技术对正常和再生24 h肝组织进行差异表达基因的筛选。结果获得10条差异序列,比对检测到其中3条与国家生物技术信息中心已表达序列标志(NCBI EST)库的序列有较高相似性,并且5条序列在肝再生过程表达上调,另5条表达下调。结论本实验结果为肝再生相关功能新基因及肝再生机制的研究提供新的有效信息。
Objective To screen differentially expressed genes in regenerative liver tissue of sharks. Methods Normal and 24 h regenerative liver tissues of Chiloscyllium plagiosum were obtained after two-thirds of partial hepatectomy( PH); 20 random primers and 2 anchor primers were used to screen differentially expressed genes in normal and 24 h regenerative liver tissues by differential display reverse transcription polymerase chain reaction( DDRT-PCR) technology. Results Ten different sequences were obtained,and 3 sequences showed high similarity to sequences of EST( expressed sequence tag) in NCBI( National Center for Biotechnology Information) by comparative detection; 5 sequences were over-expressed and another 5 sequences were down-expressed during liver regeneration. Conclusion This experiment may provide effective information for further research about the function of genes related liver regeneration and the mechanisms of liver regeneration.
