白热斯油处方搽剂及其组分对人G-361细胞活力与产生黑色素能力的影响

Effect of Bai Resi Oil Liniment and its Components on Vitality and Melanin Producing Ability of Human G-361 Cells In vitro

摘要 探讨维吾尔医白热斯油处方搽剂及其单味药、即:补骨脂、白花丹、黄芥子、黑种草子和駆虫斑鳩菊生药材水提物等通过对G-361细胞存活率、增殖以及黑色素产生的影响。对维医白癜风专用12种处方水提物进行酪氨酸酶激活实验、根据激活率(OD值)进行排序并定位。用MTT法检测不同浓度白热斯油处方搽剂及其组分药材水提物、对G-361细胞72 h细胞存活率、增值以及黑色素产生进行观测。经检测、对维吾尔医12种治疗白癜风专用药材水提物酪氨酸酶激活率超出200%的有4个专用处方、白热斯油为第3位,高于其它9种处方(P<0.001)。各药材及各浓度72 h培养细胞存活率与对照组之间相比无显著性差异(P>0.05)。72 h培养细胞增殖和黑素产生能力与对照组相比有显著性差异、分别为P<0.05和P<0.01、而且最佳浓度集中在125μg/m L和62.5μg/m L、其中后者较为稳定。白热斯油处方及其组分药材水提物在适宜浓度,对72 h培养G-361细胞有增殖作用、使黑色素含量增加、且细胞存活率正常、未发现明显的细胞毒性作用。 This paper study the effect of Uygur medicine prescription Bai Resi oil and its components,those are P.corylifolia,P. zeylanis,B. juncea,N. glanduifera,V. anthelmintica raw herbs,from their water extract and mix extract on G- 361 cell survival rate,increment and the production of melanin in vitro. First,perform a tyrosinase activation experiment on water extract of Uygur medicine vitiligo dedicated 12 kinds prescription medicines,according to the activation rate( OD value) to sort and rank. Observations of,using MTT method,the effect of different concentration water extracts of Bai Resi oil liniment and its components on 72 hr G- 361 cultured cell on cell survival,quantity increment and melanin production. After analyze,Uygur medicine vitiligo dedicated 12 kinds prescription medicines ranked by their tyrosinase activation rate from herbal water extraction,the activation rate of 4 dedicated prescriptions have exceeded 200%,and Bai Resi oil rank third,above other 9 kinds of prescriptions,differences is significant( P〈0. 001). It was found,from the survey,the treatment of Bai Resi oil prescription and its components was mainly for wet cold or phlegmatic temperaments disease,the main function for vitiligo is to increase melanin in skin lesions. Each medicinal prescription with various concentrations of 72 hr culture cell compared between control group base on their survival rate and there was no significant difference( P〈0. 05),the cell survival rate was not affected. However,base on increment and the production of melanin,the difference is significant between the control group and various concentrations of 72 h of culture cell,those are *P〈0. 05 and **P〈0. 01 respectively,the best concentration is around the 125? g / m L and 62. 5μg / m L,the later is relatively more stable concentration. Bai Resi oil medicinal prescription and its components water extract in suitable concentration has proliferation effect for 72 h cultured G- 361 cell,increased melanin content,and the cell survival rate normalliy and found no significant cytotoxicity.
