卵蛋白致敏复加臭氧暴露建立氧化应激特征性支气管哮喘SD大鼠模型研究 被引量:10

Research on SD Rats with Oxidative Stress Following Bronchial Asthma Injected by Ovalbumin and Exposed by Ozone

摘要 目的:观察清洁级雄性SD大鼠五点注射卵蛋白及臭氧暴露后肺功能、肺组织病理HE染色以及血清氧化应激指标表现,初步探索氧化应激特征性支气管哮喘SD大鼠模型的建立方法;方法:30只雄性SD大鼠随机分为3组,即空白对照A组、卵蛋白B组和卵蛋白复加臭氧C组。B、C组大鼠五点注射10%卵蛋白(OVA),A组大鼠五点注射等剂量生理盐水,每周1次,共两次;C组大鼠注射OVA后隔天进行臭氧暴露;14 d后B、C组用1%OVA连续雾化7 d诱发其哮喘发作,C组雾化同时仍隔天进行臭氧暴露,A组则给予生理盐水雾化吸入。结果:(1)B、C两组肺功能结果较A组差异有统计学意义,臭氧暴露对哮喘大鼠气道重塑有一定影响;(2)HE染色可见:B、C组支气管可见炎性细胞浸润,上皮细胞脱落;肺泡塌陷,融合;C组相比炎性浸润更为密集;(3)B、C两组血清对氧自由基的清除能力降低,与A组比较差异有统计学意义;臭氧暴露建模影响大鼠的抗氧化能力;结论:SD大鼠五点注射卵蛋白并复加臭氧暴露可成功建立氧化应激特征性支气管哮喘模型。 Objective: By observing the function of lung and HE staining and serum levels of oxidative stress in SD rats injected by ovalbumin and exposed by ozone,to explore the method of establishing the model of oxidative stress in bronchial asthma. Methods: 30 SPF female SD rats were randomly divided into three groups,blank control group A,ovalbumin group B and ovalbumin with ozone group C. Five points were injected with 10% ovalbumin( OVA) in group B and C,while five points were injected with saline in group A,once a week,for two weeks. Group C were exposed with ozone every other day after injected with OVA. After 14 days,group B and C were challenged via the airways with 1% OVA for 7 days and group C still were exposed with ozone every other day at the same time. While group A were challenged via the airways with saline for 7 days. Results:( 1) There was statistical significance between group B,C and group A in lung function and ozone exposure had an effect to reshape the airways of the rats with asthma.( 2) Biopsy pathology HE: In group B and C,it showed that bronchial visible inflammatory cell infiltration,epithelial cells fall off and Alveolar were collapse and fusion and compared to B group C inflammatory,they infiltrated more densely.( 3) The serum oxygen free radical scavenging ability of group B and C was reduced and compared to group A it was significant,P〈0. 05. By observing group C,the ozone exposure affected the antioxidant capacity. Conclusion: Five points injected with ovalbumin and exposed with ozone can successfully establish the oxidative stress model of bronchial asthma in SD rats.
