摘要
目的探讨大鼠优势半球脑缺血后基因表达谱的变化趋势。方法用行为学方法筛选出24只右利侧大鼠,制备优势半球脑缺血模型,选取双侧纹状体,用8 000个位点基因芯片扫描基因表达谱的变化。结果脑缺血组和假手术组均存在大量的低表达的EST,但各密度值范围内的EST个数不同。共有4 286个基因有差异表达,其中在基因库有定义的基因中,密度值在800以上并且Cy3/Cy5的比值>2的有368个EST,密度值在800以上并且Cy3/Cy5的比值<0.5的有79个EST,即在脑缺血组纹状体中高表达有368个,低表达有79个。其中生长因子类基因普遍表达降低,仅个别基因高表达,左侧优势侧与右侧非优势侧比较差异有统计学意义(P<0.05)。结论优势半球缺血与非优势半球缺血比较在基因表达层面,生长因子类基因表达水平更低。
Objective To investigate the trends of gene spectrum of the growth factor family following the dominant hemisphere ischemia in rat. Methods 24 right - handed male wistar rats behaviorally selected and the model of dominant hemisphere ischemia was made. 8000 spots gene chip was used to detect the gene expressions in the stratum and the sham. Results 4 286 genes were differential- ly expressed in the stratum, and were low expression in the growth factor family but seldom up - expression. The dominant hemisphere is- chemic stratum had lower expression significantly (P 〈 0.05). Conclusion The genes in the stratum express lower after the dominant hemisphere ischemia.
