摘要
目的:研究肠润通煎剂对链脲佐菌素所诱导的糖尿病大鼠结肠重构的影响。方法:40只SD雄性大鼠,30只大鼠造模后随机分为糖尿病对照组(DM组),肠润通高剂量治疗组(T1组)和肠润通低剂量治疗组(T2组),另外10只作为正常对照组(Con组)。肠润通治疗组经灌胃给药,定期测量大鼠体质量。实验结束时取出结肠中段,0.9%氯化钠溶液冲洗称湿重后,切取3个1-2mm厚的肠环用作无载荷和零应力状态测量,剩余肠段用作充压(从0-20cm水柱)实验。形态几何数据,展开角和残余应变可从无载荷和零应力状态测量中获得,结合充压肠管的压力和直径的变化,计算肠管的周向和纵向的应力和应变。结果:与Con组比较,DM,T1和T2组体质量显著降低(P<0.01)。与Con组比较,DM组的结肠肠管单位长度湿重体重比,壁厚,壁横截面积和展开角均明显增高(P<0.01),结肠壁的内外残余应变绝对值及周向和纵向刚度也明显增加(P<0.05)。T1处理能逆转糖尿病所致的形态学和生物力学变化(P<0.05),但T2处理后各项指标与DM组无显著差异。结论:肠润通煎剂(高剂量)治疗可以改善糖尿病所致的结肠形态学和生物力学重构。肠润通煎剂可能是通过改善肠道生物力学特性的重构而发挥治疗便秘的作用。
Objective: To investigate the effects of Changruntong Decoction (CRT) on the colonic remodeling in rat with diabetes induced by streptozotocin (STZ). Methods: 40 SD male rats were randomly divided into normal control group, diabetes model group (DM), high-dose Changruntong decoction group (T1) and low-dose Changruntong Decoction group (T2), with 10 rats in each group. Except for the normal control group, all rats were induced by STZ for establishing the diabetes model. The rats in the treatment groups were gavaged with different doses of Changruntong Decoction. The body weight was measured regularly. At the end of experiment, three 1-2 mm rings were cut from midpiece colon that were used for the no-load and zero-stress state tests, while the remainder of the midpiece colon were used for the step-wise distension test. The morphometric data, such as the circumferential length, the wall thickness and the opening angle were measured from the no-load and zero-stress state tests. Circumferential and longitudinal stresses and strains were computed from the pressure, length and diameter of midpiece colon after step-wise distension test. Results: The body weight in DM group, T1 group and T2 group was all significantly lower than that in control group (P〈0.01). The wet weight per unit length to body weight ratio, wall thickness, the cross-sectional wall area, opening angle in DM group were significantly higher than those in control group (P〈0.01), and the absolute values of inner and outer residual stain of colonic segments and circumference and longitudinal rigidity in DM group were higher than those in control group (P〈0.05). The changes of morphology and biomechanics in T1 group were more obvious than those in DM group (P〈0.05). While, there was no statistical difference in the changes of morphology and biomechanics between the T2 group and the DM group. Conclusion: High-dose Changruntong Decoction could partly restore the morphometric and biomechanical remodeling of colon in rats with diabetes. Changruntong Decoction could treat the constipation in clinic, the mechanism of which may be related to improve the remodeling of biomechanical properties in the intestinal tract.
