摘要
目的:研究潜阳解毒通络饮合剂对大鼠心肌细胞肥大及c-fos、c-myc mRNA的影响。方法:采用血清药理学方法,体外培养大鼠心肌细胞系,分为正常对照组、模型组、潜阳解毒通络方组、缬沙坦组、吡格列酮组。并采用甲基噻唑基四唑(MTT)法检测大鼠心肌细胞增殖抑制情况。逆转录聚合酶链反应法检测c-fos、c-myc mRNA的表达。透射电镜观察细胞超微结构。结果:血管紧张素Ⅱ(AngⅡ)作用细胞12h后,与空白组比较,c-fos、c-myc mRNA表达明显增加(P<0.05),心肌细胞增殖(P<0.05)。与模型组比较,潜阳解毒通络饮干预后,大鼠心肌细胞系增殖受到抑制(P<0.05),c-fos、c-myc mRNA表达显著下调(P<0.05)。电镜观察大鼠心肌细胞超微结构的结果表明潜阳解毒通络饮能够使线粒体和内质网扩张降低、减少溶酶体数量。结论:潜阳解毒通络饮有抑制或减少AngⅡ诱导大鼠心肌细胞肥大的作用。是通过下调c-fos、c-myc mRNA的表达。
Objective: To explore the effect of Qianyang Jiedu Tongluo Potion on the proto-oncogenes c-fos and c-myc mRNA expressions of rats with cardiomyocyte hypertrophy induced by AngⅡ. Methods: Rat myocardial cells were cultured in vitro with serum pharmacology method, which were randomly divided into normal control group, model group, Qianyang Jiedu Tongluo Potion group, valsartan group and pioglitazone group. The methyl thiazolyl tetrazolium (MTT) assay was used to detect the proliferation inhibitory of rat myocardial ceils. Reverse transcription-polymerase chain reaction (RT-PCR) was used to detect the c-los and c-myc mRNA expressions. Transmission electron microscope (TEM) was used to observe the cell ultrastructure. Results: After Ang II inducing for 12 hours, the mRNA expressions of c-fos and c-myc in all of model groups were higher than those in the normal group (P〈0.05), as well as the proliferation of myocardial cells. Compared with that in the model group, the proliferation of myocardial cells in Qianyang Jiedu Tongluo Potion group were inhibited (P〈0.05). The mRNA expressions of c-fos and c-myc in Qianyang Jiedu Tongluo Potion group were higher than those in the control group (P〈0.05). TEM results showed that Qianyang Jiedu Tongluo Potion had good effect on improving the ultrastructure of rat myocardial cell induced by Ang Ⅱ, with 10% dose Qianyang Jiedu Tongluo Potion being the best. Conclusion: Qianyang Jiedu Tongluo Potion has effect on inhibiting or reducing cardiomyocyte hypertrophy induced by Ang Ⅱ, the mechanism of which may be related to the down-regulation of c-fos and c-myc mRNA expressions.
