温郁金正丁醇提取物逆转胃癌SGC7901/VCR细胞多药耐药性的研究 被引量:9

Reversal effect of Curcuma Wenyujin n-butyl alcohol extract on multidrug resistance of gastric cancer cell SGC7901/VCR

摘要 目的:研究温郁金正丁醇提取物(CWNAE)逆转SGC7901/VCR细胞多药耐药性的作用。方法:以维拉帕米(VP)为对照采用MTT法研究无毒浓度CWNAE对SGC7901/VCR细胞多药耐药性的逆转作用。采用流式细胞仪技术观察CWNAE对VCR诱导SGC7901/VCR细胞凋亡及周期改变的影响。结果:MTT显示20、40、80μg/m L的CWNAE作用24h后均可逆转SGC7901/VCR细胞对VCR、ADM的耐药,逆转指数分别为1.51、4.07、28.95;1.59、6.29、10.34,而10μg/m L VP的逆转指数分别为21.27、95.56。流式技术显示上述浓度的CWNAE及VP可促进VCR诱导SGC7901/VCR细胞凋亡,以早期凋亡为主,并可促进VCR将细胞阻滞在G2/M期。结论:本研究证实了无毒浓度的CWNAE可以逆转SGC7901/VCR细胞的多药耐药性,促进VCR诱导SGC7901/VCR细胞凋亡及细胞周期阻滞,有剂量依赖性,80μg/m L CWNAE作用效果基本同10μg/m L VP,但毒性远比后者小。 Objective: This study mainly researched on the reversal effect of Curcuma Wenyujin n-Butyl alcohol extract (CWNAE) of multidrug resistance (MDR) in SGC7901/VCR. Methods: MTT was used to test the reversal effect of non-toxic dose of CWNAE of MDR. Flow cytometry (FC) was used to observe the influence of CENAE on apoptosis rate and cell cycle of SGC-7901/VCR induced by VCR. Results: MTT showed that CWNAE of 20, 40 and 80μg/mL could effectively reverse the VCR resistance of SGC7901/VCR, and the reversal index were 1.51, 4.07 and 28.95. It also could reverse the ADM resistance of SGC7901/VCR, and the reversal index were 1.59, 6.29 and 10.34. While the reversal index of 101μg/mL VP were 21.27, 95.56. FC showed that CWNAE could enhance the apoptosis rate of SGC7901/VCR induced by VCR and increase the ratio of cells in G2/ M stage arrested by VCR. Conclusion: This study revealed that CWNAE could reverse the MDR of SGC7901/VCR to VCR and ADM, and the mechanism was that CWNAE could increase the apoptosis rate and ratio of cells in G2/M stage of SGC7901/VCR induced by VCR. All these effects depended on the concentration of CWNAE. The effect of 80μg/mL CWNAE and 10μg/mL VP was similar, but the toxicity was far smaller than the latter. So there was a certain research value of CWNAE in the research field of reversing MDR with tumor.
