摘要
以茎瘤芥瘤茎为试材,研究了酶法释放葡萄糖测定芥子油苷的最佳条件。选用浸提温度、浸提时间、料液比进行单因素试验和正交试验研究,考查了沸水灭酶时间。结果表明:酶法释放葡萄糖测定芥子油苷的最佳条件为料液比1∶30,45℃的水浴中浸提30min;干蒸10min,沸水浴5min可致死芥子油苷酶活性。
Stems of tumorous stem mustard are used to study the optimum determination conditions for glucosinolates to release of glucose.Carry out the single factor experiment and orthogonal experiment on extraction temperature,extraction time and solid-liquid ratio to research the enzyme inactivation time with boiling water. The results show that the optimum determination conditions for glucosinolates content by glucose release assay are as follows:the ratio of material and water is1∶30,extracting for 30 min at 45 ℃in water;dry steaming for 10 min,and boiling water bath for5 min can cause a lethal to glucosinolates enzyme activity.
