摘要
本研究利用SCoT12引物对8个不同花生品种扩增产物中的一个长度多态性片段SCoT12-800进行克隆和测序分析,该序列登录号为KM200036。结果表明,该片段多态性是由于其含有一段TC简单重复序列(Simple Sequence Repeat,SSR),根据该片段序列,利用Primer 5.0软件设计出一对适合SSR分子标记检测的引物,命名为SCoT12-SSR,该引物的扩增产物大小为164-178 bp,在花生栽培种中可检测到5个等位变异,可用于花生的遗传多样性、品种鉴定、基因定位和遗传图谱的构建等研究。
In this study,one length polymorphism fragment SCoT12- 800 amplified by SCoT12 primer from 8 peanut varieties was sequenced. Its accession number was KM200036 in Gen Bank. Sequence analysis showed that the polymorphism of SCoT12- 800 was from the simple sequence repeat of TC. A new SSR molecular marker named SCoT12- SSR was designed by Primer 5.0 and its amplification product was between 164 bp and 178 bp. It could detect 5 alleles in cultivated peanuts. Thus,the marker could be used for studying genetic diversity,cultivar identification,gene tagging and genetic mapping in peanut.
