噬菌体展示技术筛选表皮生长因子受体抗原模拟表位

摘要 目的利用噬菌体12肽库进行生物淘洗,筛选表皮生长因子受体(EGFR)抗原模拟表位。方法以抗EGFR的单克隆抗体药物西妥昔单抗及尼妥珠单抗为靶分子,在噬菌体12肽库中淘选表皮生长因子受体抗原模拟表位,经过3轮淘选后,选择可与西妥昔单抗及尼妥珠单抗有不同程度结合的噬菌体,对阳性产物进行克隆及测序。结果发现17个和21个阳性噬菌体分别与西妥昔单抗及尼妥珠单抗不同程度地结合;阳性克隆测序结果分别获得了3种不同的氨基酸序列。结论初步利用西妥昔单抗及尼妥珠单抗可从噬菌体展示的随机肽库中筛选到表皮生长因子受体相关抗原表位。 Objective To screen the mimotope of epidermal growth factor using the phage display 12 peptide library.Methods With Cetuximab and nimotuzumab as the ligand,the mimotope of epidermal growth factor was screened from the phage display 12 peptide library.After three screening rounds,the positive phages which could combine the epidermal growth factor monoclonal antibody with different degrees were selected,and then cloned and sequenced.Results There were 17 and 21 phages which could combine the cetuximab and nimotuzumab with different degrees,respectively.Three different amino acid sequences were obtained by the results of those positive clones.Conclusion The epidermal growth factor receptor related antigen epitope could be obtained from phage display random peptide library using the cetuximab and nimotuzumab as the ligand.
