黄芪糖蛋白对T细胞增殖与共刺激分子表达的影响

摘要 目的:探讨黄芪糖蛋白对T细胞增殖与共刺激分子表达的影响。方法:无菌制备大鼠脾脏悬浮细胞,分离淋巴细胞,接种后加入黄芪糖蛋白共培养,采用MTT法检测淋巴细胞的增殖情况,采用流式细胞术检测淋巴细胞亚群分布。结果:MTT实验结果显示黄芪糖蛋白可显著抑制Con A刺激下淋巴细胞的增殖(P<0.05或P<0.01)。流式检测结果显示中、高剂量黄芪糖蛋白可显著降低淋巴细胞中CD3+CD54+细胞的比例(P<0.01),对CD3+CD28+细胞的比例无明显影响。结论:黄芪糖蛋白可有效抑制T细胞的增殖,同时可通过降低T细胞表面共刺激分子细胞间黏附分子-1的表达抑制T细胞的活化。 Objective: To explore the effects of glycoproteins from Astragalus membranaceus on proliferation of T cells and expression of co-stimulatory molecules. Methods: Rat spleen cell suspension was prepared and lymphocytes were isolated and cultured with different dosages glycoproteins from Astragalus membranaceus. MTT assay and flow cytometry were used to detect proliferation and subsets of T cells. Results: The results of MTT assay showed the glycoproteins significantly inhibited the prolif-eration of T cells(P〈0.05 or P〈0.01). Flow cytometry indicated the middle and high dosages glycoprotein obviously reduced the rates of CD3^+CD54^+cells(P〈0.01).Conclusion: Glycoproteins from Astragalus membranaceus can effectively inhibite the prolifera-tion and prevent the activation of T cells through inhibiting the expression of co-stimulatory molecule ICAM-1 on the surface of T cells.
