糖痛方对施万细胞凋亡及Bcl-2、caspase-3、caspase-9表达的影响 被引量:3

摘要 目的探讨中药糖痛方对高糖诱导的施万细胞凋亡的影响。方法将原代培养的施万细胞共分为对照组、高糖组、高糖加川芎嗪组、高糖加黄芪多糖组、高糖加川芎嗪和黄芪多糖混合组、高糖加糖痛方含药血清组,各组干预细胞48h后用流式细胞仪检测细胞AnnexinⅤ/PI标记的细胞凋亡率、用Western blot法和PCR法检测caspase-3、caspase-9及Bcl-2表达。结果糖痛方组细胞凋亡率与高糖组相比显著降低(P<0.05);糖痛方组细胞caspase-3、caspase-9蛋白及mRNA表达与高糖组相比显著降低(P<0.05),Bcl-2蛋白及mRNA表达与高糖组相比显著升高(P<0.05)。结论中药糖痛方对神经细胞的损伤修复作用可能是通过提高Bcl-2、下调caspase-9和caspase-3的活性表达从而抑制施万细胞的凋亡来实现的。 Objective To investigate the side effects of pain medicine sugar on Schwann cells apoptosis induced by high glucose in.Methods Primary cultured Schwann cells were divided into control group,high glucose group,high glucose plus TMP group,high glucose plus APS group,high glucose plus TMP and APS mixed group,high glucose plus TangTongfang containing serum group.After the cells in each group being intervented for 48h,the rate of apoptosis was detected by flow cytometry Annexin V/PI.The expression of caspase-3,caspase-9 and Bcl-2 was detected by Western Blot and PCR methods.Results Compared with high glucose group,the rate of apoptosis of TangTongfang group was significantly lower (P < 0.05).Compared to high glucose group,the expression of caspase-3,caspase-9 protein and mRNA of Tangtongfang group was significantly lower (P < 0.05),the expression of Bcl-2 protein and mRNA were significantly increased (P < 0.05).Conclusion Tangtongfang repairing damaged nerve cells may be by increasing Bcl-2,caspase -9 and reducing caspase-3 activity of Schwann cells to inhibit the expression of apoptosis.
