牙龈卟啉单胞菌脂多糖对牙周韧带细胞LIF mRNA表达的影响 被引量:1

摘要 目的观察牙龈卟啉单胞菌脂多糖(Porphyromonas gingivalis lipopolysaccharide,Pg-LPS)对人牙周韧带细胞(human periodontal ligament cells,HPDLCs)白血病抑制因子(leukemia inhibitory factor,LIF)表达的影响,探讨LIF与牙周病发生发展的关系。方法体外分离培养鉴定HPDLCs,取第3~5代细胞用于实验,使用不同浓度(1μg/m L和10μg/m L)的Pg-LPS刺激HPDLCs24h,实时定量反转录-聚合酶链反应检测LIF mRNA的表达。结果 Pg-LPS组LIF mRNA表达均明显高于对照组(P〈0.01),并且Pg-LPS浓度越高,LIF mRNA表达越强。结论 Pg-LPS能够诱导人牙周韧带细胞LIF表达上调,这一机制可能在牙周炎发生发展过程中发挥重要作用。 Objective To investigate the effect of Pg-LPS on the expression of LIF mRNA in the human periodontal ligament cells (HPDLCs) and to analyze the relationship with periodontitis. Methods Human periodntal tissue was ob- tained fi'om the extracted healthy premolar due to orthodontic reasons. The HPDLCs used in this study underwent three passages. Cells had been untreated or treated with lipopolysaccharide of varial viscosity (1 μg/mL and 10 μg/mL) for 24 hours. The expressions of LIF mRNA were determined by Realtime RT-PCR. Results The expression of LIF mRNA was significantly induced with the stimulus of Pg-LPS than the group of contrast. The more the Pg-LPS, the higher expression of LIF was. Conclusion Lipopolysaccharide dramatically stimulated the secretion of LIF of HPDLCs, which may plays a very important role in periodontitis.
