摘要
本文研究了金属离子对α-环糊精葡萄糖基转移酶发酵生产的影响。首先,采用单因素分析确定几种微量元素的最佳产酶浓度;其次,通过两水平析因试验确定了关键的影响因子及其最大响应区域,其分别是Mg SO4·7H2O,Fe SO4·7H2O,MnSO4·H2O;最后,通过Box-Benhnken中心组合试验建立数学模型,并采用响应面分析法获得了微量元素的最优配比参数为:MgSO4·7H2O 10.94 mmol·L-1,Fe SO4·7H2O 6.48 mmol·L-1,Mn SO4·H2O 0.51 mmol·L-1,Ca Cl2·5H2O 3 mmol·L-1。与对照组相比,酶活提高了1.4倍。
The influence of different metal ions on production of α-cyclodextrin glycosyltransferase in recombinant E.coli were discussed. Firstly, optimal concentrations of some trace elements were obtained by means of the single factor experiments. Secondly, the key factors and their largest response area were determined by the two-level factorial experiments, and the significant factors were MgSO4·7H2O, FeSO4·7H2O and MnSO4·7H2O. Lastly, the Box-Benhnken center-united experimental design and response surface methodology were utilized to optimize the ratio of the trace elements. The optimum ratio was obtained as follows: MgSO4· 7H2O 10.94 mmol·L^-1, FeSO4· 7H2O 6.48 mmol·L^-1, MnSO4·H2O 0.51 mmol·L^-1, CaCl2·5H2O 3 mmol·L^-1.
