摘要
探讨新型氨基化小分子半抗原直接包被ELISA方法检测食品中氯霉素残留的可行性。采用氨基化氯霉素直接包被在经戊二醛活化后的酶标板表面,并通过生物素-链霉亲和素放大系统进一步提高该检测方法的灵敏度。结果表明,氨基化小分子半抗原直接包被ELISA的灵敏度和最低检出限分别为12.9 ng/mL和0.7 ng/mL,CV%为1.8%~4.5%。新型氨基化小分子半抗原直接偶联ELISA具有灵敏度高,干扰少,检测步骤简便、操作安全,测定结果较准确可靠。
The novel ELISA was developed for monitoring chloramphenicol remains in food by the directly attaching small molecule hapten with amino groups to the microtiter plate surface. chloramphenicol with amino-groups was directly coated on the surface of microtiter plates by treating with glutaraldehyde solution , and biotin-streptavidin system was employed to improve the sensitivity of immunoassay. The sensitivity of the new ELISA was 12.9 ng/mL, CV%was 1.8%-4.5%. The novel amination small molecule hapten coupled directly ELISA has many advantages, such as high sensitivity, less interference, simple operation and so on.
