摘要
组蛋白去乙酰化酶抑制剂(HDACIs)调控组蛋白乙酰化酶(HAT)和组蛋白去乙酰化酶(HDACs)之间的动态平衡,控制着染色质结构和基因表达,其作用机制与表观遗传治疗学关系密切。失血性休克和复苏能诱导HAT/HDAC的比例不平衡,进一步影响细胞组蛋白或非组蛋白的乙酰化状态。HDACIs正是通过修正这种不平衡来改善组织器官的功能,具有改善失血性休克模型生存的作用。本文在收集近10年相关文献基础上,对HDACIs抗休克的作用机制及相关研究进展作一综述,为HDACIs应用基础研究提供参考。
Histone deacetylase inhibitors (HDACIs)regulate the dynamic balance between historic deacety|ases (HDAC) and histone acetyl transferases (HAT) ,which affect chromatin structure and gene expression. The mechanism of HDACIs has a close relationship with epigenetic therapy. Hemorrhage shock and resuscitation can induce the HDAC/HAT imbalance,which influences the acetylation status of histone and non-histone. HDACIs induces a biologic response during hemorrhagic shock and corrects the imbalance of HAT/HDAC,so the functions of some active organs may be inlproved. As a result ,survival effects of some animal models with hemorrhagic shock can be obtained. This review summarizes some action mechanisms and update on HDACIs against hemorrhagic shock in the recent ten years which may provide some referenc e for the basic applied research about HDACIs.
