摘要
以去甲万古霉素为手性选择剂,采用4,4-二苯基甲烷异氰酸酯作为间隔臂,将其键合到氨化的单分散交联聚甲基丙烯酸环氧丙酯微球上,合成了去甲万古霉素手性固定相。该键合固定相在极性有机相模式下,成功地实现了五种手性药物对映体的分离,详细考察了流动相组成、酸碱添加剂用量、温度和流速等对药物对映异构的拆分影响,并初步探讨了其手性识别机理。
An antibiotic named norvancomycin was used as a new chiral selector, with 4,4-two phenyl methane diisocyanate as spacer arm to synthesis on the monodisperse poly ( glycidylmethacrylate-co-ethylenedimethacrylate ) beads for the preparation of norvancomycin-bonded chiral stationary phase ( NVC-CSP ) . The possible mechanisms of chiral discrimination were also discussed based on the enantioseparation of versatile structures of chiral compounds under different chromatographic modes. The mechanism of chiral separation was preliminarily studied.
