摘要
目的研究双相障碍Ⅰ型患者外周血DNA甲基化水平的变化。方法提取20名双相障碍Ⅰ型患者和20名健康对照者的外周血单个核细胞的DNA,通过重亚硫酸氢钠测序方法测量疾病组和对照组的DNA甲基化水平,用杨氏躁狂评定量表评定所有参与者的躁狂程度,并用Pearson相关分析方法进一步研究DNA甲基化水平和躁狂程度的关系。结果双相障碍Ⅰ型患者外周血DNA甲基化水平明显升高,而且其升高程度与躁狂程度呈正相关关系。结论 DNA甲基化水平可能与双相障碍发病有关。
Objective The purpose of this study was to explore the peripheral DNA methylation in bipolar patients during manic phase.Methods DNA was extracted from peripheral blood leukocytes of 20 euthymic bipolar patients type I and 20 matched healthy controls.Percent of global genome DNA methylation was measured using the cytosine-extension method.The Young Mania Scale was used to evaluate the severity of mania.And then Pearson correlation was used to explore the relationship between DNA methylation and the severity of mania.Results Compared with the healthy subjects, the manic patients showed increased DNA methylation in peripheral blood, and DNA methylation was positively correlated with Young Mania Rating Scale scores in bipolar disorder patients.Conclusions DNA methylation may play an important role in the etiology of the disorder.
