摘要
目的探讨Ascl2与MicroRNA-200家族在结肠癌(colorectal cancer,CRC)组织及癌旁组织中的表达及二者的相关性。方法采用实时定量PCR检测50例结肠癌患者的结直肠癌组织及其癌旁组织中Ascl2与MicroRNA-200家族表达,用SPSS 18.0统计软件进行统计分析。结果 Ascl2在癌组织中的表达显著高于癌旁组织(P<0.01),除了MicroRNA-429在癌组织中的表达与癌旁组织相比无统计学差异(P=0.2139),MicroRNA-200家族成员在癌组织中的表达都显著低于癌旁组织(P<0.05);且Ascl2和MicroRNA-200家族成员在癌组织中表达都呈负相关(r<-0.5,P<0.05)。结论发现与正常结直肠黏膜组织相比,Ascl2在结直肠癌组织中表达增高,MicroRNA-200家族表达降低,它们在结直肠癌组织中的表达呈现负相关。
Objective To investigate the expression of Ascl2 and microRNA-200 family members in colorectal cancer and their peri-cancerous mucosa tissues and determine their correlations. Methods The expression of Ascl2 mRNA and microRNA-200 family members in 50 colorectal cancer samples and their pericancerous mucosa was detected by real time-PCR. Statistical analysis was performed by SPSS 18 statistical software. Results The mRNA expression level of Ascl2 was significantly higher in cancerous samples than the corresponding peri-cancerous mucosa( P 〈 0. 01),while the levels of microRNA-200 family members were significantly lower in cancerous samples than the peri-cancerous mucosa( P 〈 0. 05),except that no difference was found in microRNA-429 expression between the 2 tissue samples( P = 0. 2139). The expression level of Ascl2 was negatively correlated with those of microRNA-200 family members( r 〈- 0. 5,P 〈 0. 05).Conclusion The expression level of Ascl2 is increased,while those of microRNA-200 family members are decreased in colorectal cancer tissues when compared with the corresponding peri-cancerous tissues. And the expression levels are negatively correlated.
