摘要
为探讨脂联素(adiponectin,APN)m RNA表达与新疆维吾尔族肥胖及2型糖尿病的相关性。将样本分为3组:正常组(18.5 kg/m2≤BMI≤24 kg/m2)50例,肥胖组(BMI≥28 kg/m2)48例,2型糖尿病组(餐后2 h血糖≥11.1mmol/L空腹血糖≥7.0 mmol/L)16例,腹部择期手术时取其网膜脂肪组织,采用实时定量RT-PCR(real-time quantitative reverse transcription polymerase chain reaction)对上述3组样本中的脂联素m RNA表达进行检测。采用SPSS17.0进行统计学分析。结果显示,肥胖及糖尿病组个体体重、腰围、臀围、腰臀比及体质指数均显著高于正常对照组,差异有统计学意义(P<0.01)。肥胖组男女性别间差异有统计学意义(P<0.01)。比较各组脂联素m RNA相对拷贝数后发现,正常对照组显著高于肥胖及糖尿病组,差异均有统计学意义(P<0.01)。同时肥胖组高于糖尿病组,但差异无统计学意义(P=0.753)。腹部脂肪组织脂联素m RNA相对拷贝数与BMI显著负相关(r=-0.334,P=0.045),与腰围显著负相关(r=-0.053,P=0.047),与血糖水平显著负相关(r=-0.129,P=0.017)。由此可知,腹部脂肪组织中脂联素m RNA的低表达是新疆维吾尔族肥胖及2型糖尿病发生的危险因素之一。
To discuss the relationship between the mRNA display of the Adiponectin (APN) and Xinjiang Ugyr people's obesity as well as the type-2 diabetes mellitus(T2DM).Three groups of the fat samples were collected:50 samples for normal group (18.5 kg/mE≤BMI≤24 kg/m2); 48 samples for obesity group (BMI≥28 kg/m2);16 samples for T2DM group (after meal blood sugar≥ 11.1 mmol/L and limosis blood sugar 〉1 7.0 mmol/L),then detected all mRNA displays according to the RT-PCR and analyzed by SPSS17.0.All parameters in terms of the weight, the waist size,the hip size,waist-to-hip ratio and the physique index from the obesity and T2DM groups were obviously higher than the normal group was significantly higher than the obesity and T2DM group (P〈0.01).It was found that the diffemece of relative copy data of the Adiponectin mRNA,from all groups was statistical ly significant (P〈0.01). Meanwhile, the data of the adipose cytokines from the obesity group was also higher than that from the T2DM groups,however,there was no statistical significance (P=-0.753). The relative copy data of the Adiponectin mRNA in Abdominal adipose tissue was negatively correlated to the BMI (r=-0.334,P=0.045), the waist size (r=-0.053,P=-0.047) and the blood sugar level (r=-0.129,P=-0.017).Low expression of adiponectin mRNA of the abdominal adipose tissue is one of the risk factors of Uygur people obesity and T2DM.
