摘要
目的建立灭菌定性试验中和剂鉴定方法。方法按照评价灭菌效果的试验体系和试验过程,直接在中和剂肉汤、中和产物肉汤中加入试验菌的方法进行实验研究。结果含500 mg/L硫代硫酸钠肉汤及其与≤3 000 mg/L过氧乙酸作用后形成的中和产物肉汤对枯草杆菌黑色变种芽孢的恢复生长无影响。含2 000 mg/L硫代硫酸钠肉汤及其与≤5 000 mg/L过氧乙酸作用后形成的中和产物肉汤可恢复大量枯草杆菌黑色变种芽孢的生长,但对少量的枯草杆菌黑色变种芽孢的恢复生长有影响。含≥3 000 mg/L硫代硫酸钠肉汤与任何浓度的过氧乙酸作用形成的中和产物肉汤对枯草杆菌黑色变种芽孢的恢复生长均有影响。结论按照评价灭菌定性效果的试验体系和试验过程建立的灭菌定性中和剂鉴定方法,是一种科学、合理、准确的中和剂鉴定方法。灭菌定性试验中,应严格鉴定中和剂的有效性和对试验菌恢复生长的影响。
Objective To establish the neutralizer identification method for qualitative test of sterilization. Methods Based on the test system and process to evaluate the effect of qualitative sterilization, bacteria were added directly into the neutralizer broth and neutralizing product broth respectively. Results The neutralizing product broth containing 500 mg/L sodium thiosulfate broth with ≤3 000 mg/L peracetic acid did not affect the growth of Bacillus subtilis var. niger spores. However, the neutralizing product broth containing 2 000 mg/L sodium thiosulfate with ≤5 000 mg/L peracetic acid could recovered the growth of spores, but affected the recovery growth of few spores. Moreover, the neutralizing product broth containing ≥3 000mg/L sodium thiosulfate with any amount of peracetic acid affected the recovery growth of the spore. Conclusion The method based on the test system and process to evaluate the effect of qualitative sterilization is a scientif- ic, reasonable, accurate neutralizer identification method. In the experimental process, the effectiveness of neutralizer and the affect of recovery growth of test organisms could be measured rigorous.
