摘要
目的研究Hp感染对胃黏膜FOXP3、TGF-β1、IL-10表达的影响。方法采用原位杂交和ELISA法检测Hp感染与未感染患者胃黏膜活检组织中FOXP3m RNA、TGF-β1m RNA的表达和IL-10的含量;用ELISA法检测胃黏膜上皮细胞GES-1与Hp共培养后的TGF-β1、IL-10含量。结果 (1)Hp感染阳性患者胃黏膜组织中FOXP3m RNA、TGF-β1m RNA的阳性表达率及IL-10的含量较Hp感染阴性患者明显增高(分别为P<0.05、P<0.05和P<0.01);(2)Hp感染阳性患者胃黏膜组织FOXP3m RNA的表达与TGF-β1m RNA和IL-10的表达呈正相关(分别为P<0.05和P<0.01);(3)胃黏膜上皮细胞GES-1与Hp共培养1h后,TGF-β1及IL-10的表达量均较阴性对照组明显增高(P<0.05)。结论 Hp感染可上调胃黏膜局部CD4+CD25+FOXP3+Treg细胞,诱导胃黏膜细胞分泌TGF-β和IL-10,从而抑制胃粘膜局部免疫,促进Hp的免疫逃逸和持续性感染。
Objective To explore the effect of Helicobaeter pylori (H.pylon) infection on the expression of FOXP3, TGF-β1 and IL-10 in gastric mucosa. Methods The hybridization in situ and ELISA were respectively used to determine theexpression of Foxp3 mRNA, TGF-ISlmRNA and IL-10 in gastric mucosa. The expression of TGF-β1 and IL-10 in gastricepithelial cells infected with H. pylori were detected by ELISA. Results FOXP3mRNA, TGF-β1mRNA and IL-10 wereincreased in gastric mucosa infected with H.pylori compared to uninfected ones (P〈0.05, P〈0.05 and P〈0.01, respectively).Furthermore, the expression of TGF-lmRNA and IL- 10 were correlated with the expression of FOXP3mRNA in gastricmueosa infected with H.pylori (P〈0.05 and P〈0.01, respectively). In addition, the co-incubation of GES-1 with H.pylori led toincrease of the expression of TGF- β1 and IL- 10 protein (p〈0.05). Conclusion H.pylo infection could affect theexpression of FOXP3, TGF-β1 and IL-10 in gastric mueosa which probably contributed to bacterial persistence and possiblyalso to gastric tumor progression.
