TMEM16A在口腔癌前病变和鳞癌中的表达及意义 被引量:1

Expression and significance of TMEM16A in oral precancerous lesions and squamous cell carcinomas

摘要 目的检测跨膜蛋白16A(TMEM16A)在口腔癌前病变组织及口腔鳞癌组织中的表达与分布情况,分析其在口腔癌变机制中的作用。方法采用免疫组织化学染色法检测10例正常黏膜组织、10例上皮单纯性增生、30例上皮异常增生和45例口腔鳞癌标本中TMEM16A的表达情况。结果 TMEM16A在正常黏膜组织上皮中为阴性表达,在口腔癌前病变组织出现不同程度的表达,且随着上皮异常增生程度增加,TMEM16A蛋白表达逐渐增强。TMEM16A在口腔鳞癌中表达显著高于口腔癌前病变,在口腔鳞癌I、II、III级中,TMEM16A的表达差异无统计学意义(χ2=1.74,P>0.05)。结论 TMEM16A在不同级别口腔癌前病变及鳞癌中表达的差异提示其可能在口腔鳞癌发生早期有重要作用。 Objective To investigate the expression and distribution of TMEM16A in cases with oral squamous cell carcinomas (OSCCs) and oral precancerous lesions. Methods The expression of TMEM16A in samples of cases with oral precancerous lesions and OSCCs, including 10 normal oral mucosa cases, 15 epithelial hyperplasia cases, 30 epithelial dysplasia cases ((12 mild, 9 moderate, and 9 severe), and 45 OSCCs cases (21 well differentiated, 16 moderately differentiated, 8 poorly differentiated) were immunochemically determined. Results TMEM16A expression was not detectable in the normal squamous epithelium. Variable membraneous/cytoplasmic staining was observed in oral precancerous lesions and OSCCs. Statistical analysis revealed a significant increase in TMEM16A expression among cases with epithelial hyperplasia, mild epithelial dysplasia, moderate epithelial dysplasia, and severe epithelial dysplasia. TMEM16A expression was significantly increased in cases wirh OSCCs than that with oral precancerous lesions without showing significant difference among cases with well differentiated, moderately differentiated, and poorly differentiated OSCCs (2=1.74, /9〉0.05). Conclusions The differences in the immunoexpression of TMEM16A between different grades of cases with oral precancerous lesions and OSCCs suggest TMEM16A may participate in the multistep process of oral carcinogenesis since its early stages.
