MUC4、MUC16在卵巢上皮癌细胞系SKOV-3、OVCAR-3、CAOV-3和组织中的表达及临床意义 被引量:2

摘要 目的 :研究MUC4、MUC16在卵巢上皮癌细胞系SKOV-3、OVAR-3、CAOV-3和卵巢上皮癌组织的表达及与临床病理特征关系。方法:1培养卵巢癌SKOV-3、OVAR-3、CAOV-3细胞系,行免疫细胞化学SABC法和Western blot观察MUC4、MUC16蛋白表达;2对临床卵巢癌组织采用免疫组织化学SP两步法检测正常卵巢组织和肿瘤组织中MUC4、MUC16表达与临床病理特征关系。结果:1经免疫细胞化学和Western blot研究发现MUC4在人卵巢癌SKOV-3和OVCAR-3细胞中不表达,而在CAOV-3细胞中有表达;MUC16在人卵巢癌OVCAR-3细胞中表达量最高,其次是CAOV-3细胞,在SKOV-3细胞表达量最低;2在正常卵巢组织和卵巢良性肿瘤中,MUC4和MUC16表达量均较低。但在卵巢交界性肿瘤和卵巢癌中,MUC4和MUC16表达非常明显;3 MUC4在卵巢浆液性和黏液性上皮癌中表达无差异(86.36%vs 75.00%),而MUC16在浆液性卵巢癌中阳性表达率(81.81%)明显高于黏液性卵巢癌(25.00%)(P<0.05)。MUC4在早期卵巢癌表达偏高,达91.67%,而MUC16在晚期卵巢癌表达(85.70%)明显高于早期(50.00%)。结论:不同卵巢癌细胞MUC4和MUC16表达不同;联合MUC4和MUC16的检测有助于卵巢癌的病理分型、临床诊断及预后的判断。 Objective:To investigate relationship between expression of MUC4 and MUC16 and clinical pathological characteristics in ovarian cancer cell line SKOV-3,OVAR-3,CAOV-3 and epithelial ovarian cancer(EOC) tissues. Methods: ①Ovarian cell line SKOV-3,OVAR-3 and CAOV-3 were cultured,and then immunocytochemistry and Western Blot analysis were taken to detect the protein expression of MUC4 and MUC16; ②Ovarian normal and epithelial cancer tissues were collected. IHC was used to detect the protein expression of MUC4 and MUC16 in ovarian normal and cancer tissues. Results:①The immunocytochemistry and Western blot analysis showed that the expression of MUC4 protein was not detected in SKOV-3 and OVCAR-3 cell lines,but it was observed in CAOV-3. While,the expression of MUC16 protein was detected in three ovarian cancer cell lines. Moreover,the level of MUC16 was highest in OVCAR-3 cell line,second in CAOV-3 and the lowest in SKOV-3. The differences were statistically significant(P〈0.05);②The positive expression of MUC4 and MUC16 was low in normal ovarian tissue and benign ovarian tumor,but was high in border line ovarian tumor and EOC; ③the positive expression rate of MUC4 and MUC16 among different subtypes including serous tumor(86.36%,75%);mucinous tumor was(75%,25%) respectively. The positive expression rate of MUC4 and MUC16 at different tumor stages was including early stage(91.67%,50%) and advanced stage(75%,85.7%). Conclusion:① There were different expressions of MUC4 and MUC16 in ovarian epithelial cancer cell lines; ② MUC4 alone or together with MUC16 may be as a good diagnosis andprognosis marker for EOC.
