摘要
目的 :寻找特发性膜性肾病(idiopathic membranous nephropathy,IMN)患者外周血单个核细胞小核糖核苷酸(micro RNA)的差异性表达以及发生碱基突变的难易度。方法:运用高通量测序技术分别对IMN患者和健康对照者(NC)的micro RNA进行测序,构建IMN与NC组之间micro RNA差异性表达谱。并对两组的micro RNA进行碱基编辑分析,寻找发生了碱基突变的micro RNA,比较IMN与NC组micro RNA碱基突变的难易程度。结果:通过构建表达谱,找到了has-mi R-208b、has-mi R-195-3p、has-mi R-23b-5p、has-mi R-95、has-mi R-503、has-mi R-449a、has-mi R-486-3p与has-mi R-27 8个micro RNAs最具有差异性表达。2组共同表达的41个micro RNA中,IMN比NC更容易发生碱基编辑而引起碱基突变。结论:IMN患者与健康对照者的micro RNA存在着差异性表达,差异性表达的micro RNA特异性很高,可以作为深入研究IMN发病机制的靶点。IMN患者相比健康对照者更容易发生micro RNA碱基编辑引起碱基突变,这些发生碱基突变的micro RNA可能用于解释IMN的病因基础。
Objective:To identify micro RNA with different expression and the ease on base mutation based on peripheral blood mononuclear cell from idiopathic membranous nephropathy(IMN) patients. Methods:We used the high-throughput sequencing technology to establish the differently expressed spectrum of micro RNA of IMN patients and healthy people(the control group),respectively. We found out the significantly different expression of micro RNA. We also analyzed the base edited in the micro RNA to find out the micro RNA which happened in base mutation, to compare the micro RNA between the two groups which in common to evaluate the ease of base mutation. Results:After the creation of micro RNA different expression profile,we found out eight micro RNAs,which were most significantly different expression,including has-mi R-27,has-mi R-208 b,has-mi R-195-3p,has-mi R-23b-5p,has-mi R-95,has-mi R-503,has-mi R-449 a and has-mi R-486-3p. In the common micro RNAs between the IMN and the NC groups,there are forty one micro RNAs,which had the greater opportunity to have the base edited in the IMN group than in the NC groups. Conclusion:The micro RNA expression between the IMN and the NC groups showed significant differences,and the significantly different expression micro RNAs were special that could be used as target to further research the pathogenesis of IMN. The micro RNAs of IMN patients were more easily to have base edited than that of NC group,and the micro RNAs with edited base coulbe possibly related to the pathological mechanism of IMN.
