摘要
抗体是生物传感器中主要的检测探针,因此抗体的固定对传感器性能有重要影响。高效固定抗体并最大限度地保持抗体的活性,有利于提高传感器的灵敏度、稳定性和特异性。生物传感器中常用的抗体固定方法有共价偶联法、直接吸附法、生物素-亲和素法、金属螯合法和A蛋白法等。共价偶联法固定抗体有较高的稳定性、重现性与较低的非特异性结合率,是抗体固定中最常用方法之一。利用蛋白质工程在抗体分子的恒定区引入功能蛋白,有利于抗体的定向固定。
Antibody immobilization plays a key role in immuodetection in biosensors. Immobilizing the antibody effectively can help to improve the sensitivity, stability and specificity of the biosensor. In this paper, we investigate the methods used in antibody immobilization, including covalent immobilization, physical adsorbing, biotin-avidin immobilization, metal chelate immobilization, protein A immobilization and so on. Covalent immobilization stands out for its high stability and low non-specificity, becoming one of the most common methods for antibody immobilization. Protein engineering introduces functional protein to the constant region of the antibody which is helpful to the orient immobilization.
