REDD1在胎盘组织中的表达及意义

摘要 目的探讨发育及DNA损伤反应调节基因1(regulated in development and DNA damage responses 1,REDD1)在人胎盘组织中的表达及其意义。方法选择早孕绒毛组织(6~10周)、中孕(24~28周)及足月晚孕(38~39周)胎盘组织各8例,采用免疫组织化学染色技术,Western blot杂交以及实时荧光定量聚合酶链反应(polymerase chain reaction,PCR)检测REDD1 mRNA及蛋白在不同孕周胎盘组织中的表达。结果 REDD1蛋白表达于胎盘滋养细胞的细胞质中,随着孕周增加,REDD1在mRNA及蛋白在胎盘组织中的表达呈现下降趋势。结论 REDD1可能早期参与了滋养细胞侵袭的过程。 Objective To explore the significance of the expression of regulated in development and DNA damage responses 1( REDD1) in human placentas. Methods The study involved placental samples from different gestational weeks: early villous tissues( gestational age: 6 ~ 10 weeks),second trimester( gestational age: 24 ~ 28 weeks) and third trimester( gestational age: 38 ~ 39weeks) placentas,with 8 cases in each group. REDD1 protein in the placenta was examined and located by immunohistochemistry and Western blot. Additionally,REDD1 mRNA expression level was determined by relative quantitative real time fluorescence PCR.REDD1 protein and mRNA expression levels were compared between the three groups. Results REDD1 protein was predominantly observed in the cytoplasm of villous cytotrophoblasts and trophoblasts. Compared with early pregnancy,expression of REDD1 protein and mRNA level decreased in middle and late pregnancy. Conclusion REDD1 may be early evolved in procedure of trophoblasts invasion.
