基于指纹图谱与一测多评法相结合的五味子质量评价 被引量:9

Quality Evaluation of Schisandra chinensis Based on Combination of Fingerprint and QAMS

摘要 建立了高效液相色谱指纹图谱及一测多评法同时测定五味子中的6个木脂素类成分。采用Lichrosphere C18色谱柱,以乙腈∶水为流动相进行梯度洗脱,检测波长217 nm。测定了12批五味子样品的指纹图谱,相似度大于0.990,建立了指纹图谱共有模式,共标定了24个共有峰。采用已建立的一测多评法,以五味子醇甲为内参物,通过相对保留值定位待测组分五味子醇乙、五味子酯甲、五味子甲素、五味子乙素和五味子丙素,外标法测定五味子醇甲的含量,并通过相对校正因子计算其他5种待测组分的含量。五味子醇甲在0.09~1.62?g范围内线性关系良好,回收率为97.78%,RSD为2.32%。 An HPLC fingerprint of Schisandra chinensis was established, and a quantitative analysis of multicomponents by single marker(QAMS) method was developed for the simultaneous determination of six lignanoids in S. chinensis. A Lichrosphere C18 column was used, with the mobile phase of acetonitrile∶water by gradient elution, at the detection wavelength of 217 nm. Fingerprints of 12 batches of S. chinensis were determined and the similarities were over 0.990. The common mode of fingerprint was established and 24 common peaks were demarcated. By established QAMS method, schizandrol A was selected as the internal reference, then peak positions of schizandrol B, schisantherin A, deoxyschizandrin, schizandrin B and schizandrin C were definited by relative retentions, respectively. Content of schizandrol A in 12 batches of samples was determined by regression method and contents of the other components were calculated by relative correction factors(RCFs), respectively. It was linear for schizandrol A in the range of 0.09-1.62 ?g. Its recovery was 97.78%, with RSD of 2.32%.
