摘要
本试验通过测定EPP和NEPP对IL-1β和IL-6mRNA表达的影响,确定蓝莓可萃取多酚(EPP)及不可萃取多酚(NEPP)是否具有抗炎作用.结果表明,蓝莓中EPP和NEPP均能抑制细胞中IL-1β、IL-6 mRNA的表达,EPP的抑制效果要优于NEPP.当培养时间为48 h时,EPP和NEPP的添加浓度为10~ 200μg·mL-1对IL-1β和IL-6 mRNA抑制作用明显.当EPP的添加浓度为100μg·mL-1时,在12 ~48 h培养时间内,对IL-1β和IL-6 mRNA抑制作用明显;而100μg· mL-1 NEPP处理组中,在48h时对IL-1β和IL-6 mRNA抑制效果明显.这表明从蓝莓中的NEPP同EPP一样可通过抑制IL-1β、IL-6 mRNA的表达表现出明显的抗炎作用,对今后蓝莓在医药保健方面研究奠定基础,不可萃取多酚的研究为多酚研究提供了新的方向.
The effects of EPP and NEPP from blueberries on mRNA expression levels of IL - 1β and IL -6 in LPS- induced RAW 264.7 cells were studied in the present paper. The results showed that the mRNA expression levels of IL - 1β and IL - 6 in macrophages were decreased by EPP and NEPP, and the inhibition effect of EPP was better than that of NEPP. The strong inhibition effect on IL- 1β and IL-6 mRNA expression was detected in 10 -200 μg·mL-1 EPP or NEPP, at 48 h. In the treatment of 100 μg· mL -1 EPP, the significant inhibition effect on IL- 113 and IL- 6mRNA expression was detected at 12-48 h, and the significant inhibition effect on IL-6mRNA expression was detected at 48 h in the treatment of 100 μg·mL -1 NEPP. It is suggested that EPP and NEPP from blueberries have significant anti- inflammatory effect by inhibiting expression levels of IL - 1β and IL - 6 mRNA in macrophages.
