HPLC法测定枸杞鲜果中主要类胡萝卜素组成 被引量:19

摘要 建立了枸杞果实中5种主要类胡萝卜素新黄质、叶黄质、玉米黄素、β-隐黄质及β-胡萝卜的高效液相色谱分析方法,分析测定了7个不同品种枸杞果实中5种主要类胡萝卜素含量。样品经四氢呋喃提取、皂化8 h后分析测定,采用YMC-C30柱(4.6 mm×250 mm,5μm)分离,流动相A相:V((0.1%三乙胺)甲醇)︰V(水)=75︰25,B相:V(丙酮)︰V(二氯甲烷)︰V(甲醇)=70︰5︰25梯度洗脱,流速1 m L/min,柱温25℃,在波长450 nm处检测。5种类胡萝卜素的平均回收率为87.5%~91.5%,最低检出限为0.000 7μg/μL^0.006 2μg/μL。方法的准确度和精密度都很高,可用于枸杞鲜果类胡萝卜素的定量分析。 A high–performance liquid chromatagraphy method was developed for simutaniously determine 5 main carotenioids in fruits of Lycium bararum. Five main carotenoids in fruits of Lycium barbarum of seven varieties were identified and quantified. Samples were extracted with THF and saponified for 8 h followed by HPLC analysis. By employing a YMC-C30(4.6 mm×250 mm, 5 μm) column, 5 main carotenois were separated using a gradient mobile phase of V((0.1% triethylamine) methanol)︰V(water)=75︰25, and V(actone)︰V(dichloromethane)︰V(methanol)=70︰5︰25, with flow rate at 1 m L/min, column temperature at 25 ℃ and detection wavelength at 450 nm. The average recovery of 5 main carotenioids was 87.5%-91.5%, the limit of detection was 0.000 7 μg/μL-0.006 2 μg/μL. This method was reliable and accurate,and it can be used for quantitative analysis of carotenioids in Lycium bararum fresh fruits.
