摘要 目的主要研究镉对卵泡颗粒细胞凋亡和DNA完整性的影响。方法通过向培养液中添加不同浓度的Cd Cl2使小鼠卵泡颗粒细胞处于Cd暴露的条件下,采用流式细胞术、DNA Ladder和台盼蓝染色的方法检测细胞的凋亡和DNA完整性。结果低浓度(5μM)的Cd Cl2处理细胞24 h可引起小鼠细胞的大量死亡,高浓度(20μM和50μM)Cd Cl2处理颗粒细胞12 h即可引起细胞的大量死亡;流式细胞术结果显示,Cd Cl2处理颗粒细胞后,随着Cd浓度的增加,无论凋亡早期、凋亡后期和死细胞所占比例都会极显著的增加(P<0.01);Cd引起的细胞凋亡过程中伴随着DNA的片段化。结论 5μM的镉即可引起小鼠颗粒细胞的凋亡,进而可能影响小鼠卵巢的正常正理功能。 Objective To study the effect of cadmium on apoptosis of mice granulosa cells and DNA integrity. Method Mice granulosa cells were exposed to cadmium ( Cd) by adding CdCl2 of different concentrations into the culture medium. Apoptosis and DNA integrity of granulosa cells was detected by using flow cytometry, DNA Ladder and trypan blue staining. Results Lower concen-tration (5 μM) CdCl2 treated cells for 24 h or higher concentration (20 μM and 50 μM) CdCl2 treated granulosa cells for 12 h could cause massive death of cells, flow cytometry results showed that with the increase of Cd concentration, the proportion of early apoptosis, late apoptosis, and dead cells was significantly increased after the treatment with different concentration of CdCl2 (P〈0. 01), and mean-while, DNA fragmentation was also found in the process of apoptosis of granulosa cells induced by Cd. Conclusion 5μM concentration of cadmium can induce apoptosis of granulosa cells in mice, which might further affect the normal function of mouse ovaries.
