摘要
半薄切片的制备是一种常用制样手段,图像清晰度、分辨率远优于石蜡切片,视野大于超薄切片,有利于获得高质量的光镜图像,同时也是电镜超薄切片技术中一种有效的定位方法。尽管一些关键技术如固定、切片、染色有其特点和难点,但是已经成为现在结构观察中不可缺少的手段之一,在电镜观察中发挥着越来越大的作用和优势。本文从固定液的选择、切片的制备、脱树脂和染色几个方面进行探讨,为找到最好的制片方法提供参考。
Preparation of semi-thin sections is a common sample preparation method. The image is clear, the resolution is far superior to paraffin, the vision is greater than the uhrathin sections, and it is conducive to high-quality optical microscope image, hut also is a kind of effective positioning methods for the electron microscope in an ultra-thin sectioning technique. Although some key technologies such as fixing, sectioning and staining have their own characteristics and difficulties, but it has become one of the structures observed in the indispensable means and now plays an increasingly important role in advantages of electron microscopy. This paper discusses the choice of fixative prepared slices, removing resin and staining aspects, to provide a reference for the producers to find the best method.
