摘要
目的 探讨硫芥中毒动物淋巴细胞的损伤特点及中毒早期白细胞介素的浓度变化作为硫芥毒性损伤诊断的可能性 .方法 分离大鼠脾淋巴细胞培养 ,经 10~ 10 0 0 μmol·L- 1 芥子气染毒 30 min,以单细胞凝胶电泳法 (SCG )检测DNA损伤 ;同时测定硫芥中毒大鼠血液 IL- 2和 IL- 6浓度 .结果 中毒后淋巴细胞 DNA迁移率和受损细胞百分率呈时间和剂量依赖关系 (r=0 .6 3,P<0 .0 1) ;培养液中 IL- 2和IL- 6浓度明显降低 ,IL- 6含量为 (46 8± 16 ) μg· L- 1 ,4 h开始下降 ,72 h降至 (177± 16 )μg· L- 1 (P<0 .0 1) ,12 0 h有所回升 ,但仍然明显低与对照 (P<0 .0 1) .其中 IL- 2几乎难以检出 .结论 硫芥中毒早期机体免疫系统受到明显抑制 ,SCG检测
AIM To investigate the characteristics of the lymphocytes DNA damage in rats treated with sulfur mustard. METHODS Spleen lymphocytes of rats having been exposed to 10~1000 μmol·L -1 mustard gas at 37 ℃ 30 min. Were isolated and DNA damages in lymphocytes at 0, 4, 24 and 72 h postexposure were measured with single cell gelelectrophoresis assay (SCG). The concentrations of IL 2 and IL 6 were also measured after the rats were intoxicated by Mus tard gas. RESULTS There was a time and dose dependent relationship between mustard gas, and DNA migration and the percentage of DNA damaged in lymphocytes, and a significant increase in DNA damage at 10 -3 mol·L -1 and 10 -4 mol·L -1 was observed after 4 h postexposure compared with the control group. The interleukin declined release from (468± 16) μg·L -1 to (177±16) μg·L -1 of the lymphocytes at the early stage following mustard gas challenged (r= 0.63, P <0.01). CONCLUSION SCG may be an indicator of cytotoxicity resulting from mustard gas, which induces immunorepressive reaction in rats.
