摘要
针对副溶血性弧菌(Vibrio parahaemolyticus)在低温条件下容易进入不可培养状态,难以保藏的问题,通过平板菌落计数法对副溶血性弧菌的4℃冰箱保藏条件进行了探讨。结果表明,NaCl对副溶血性弧菌4℃保藏有保护作用,其中以45 g/L NaCl保护效果最佳,而且45 g/L NaCl不影响副溶血性弧菌的生长。将副溶血性弧菌细胞浸于45 g/L NaCl溶液或以含45 g/L NaCl的培养基培养后直接于4℃保藏,保藏90 d,活细胞数仅由109下降到106。
Considering that Vibrio parahaemolyticus is easy to be viable but nonculturable at low temperature,the preservation conditions of V.parahaemolyticus at 4 ℃ were investigated with plate count method.The results showed that NaCl was very helpful for V.parahaemolyticus preservation at 4 ℃.The optiomal concentration of NaCl for the V.parahaemolyticus protection was 45 g/L.45 g/L of NaCl had no negative effects on the growth of V.parahaemolyticus.When preserved V.parahaemolyticus at 4 ℃ by immersed the cells in 45 g/L of NaCl or directly used the liquid cultures cultivated with the liquid media containing 45 g/L of NaCl,the number of culturable cells were slightly decreased from initial 109 to 106 after 90 d.
