桑黄菌丝多糖的提取及多糖成分分析 被引量:8

Optimizing the Extraction Conditions of Polysaccharides from Phellinus Igniarius Mycelium and Analysis of the Components of Polysaccharides

摘要 以桑黄(Phellinus igniarius)菌丝体为材料,探索热水提取法提取桑黄菌丝体多糖的工艺,并分析桑黄多糖的主要组成成分。试验在100℃的恒温下,以多糖提取率为考察目标,分别对提取料液比(m/V,g:mL)和提取时间进行考察,利用硅胶薄层分析的方法,对桑黄多糖的成分做初步测定。热水提取的最优工艺为在水提温度为100℃条件下,料液比1∶45、浸提时间3.5 h,此时桑黄粗多糖提取率为3.99%;桑黄菌丝体多糖的单糖组成初步确定有D-葡萄糖、D-半乳糖、L-阿拉伯糖和D-乳糖。 To optimize the hot water extraction conditions of polysaccharides from Phellinus igniarius,mycelium was used as material and the main components of its polysaccharides were analyzed.Using the extraction yield of total polysaccharides as target,the orthogonal test of three factors and three levels were made under the temperature of 100 ℃.The three factors were extraction ratio of material to liquid,extraction time,and blank,extraction ratio of material to liquid.By the method of paper chromatography and silica gel thin layer,the optimized condition of hot water extraction method were under 100 ℃ of the water temperature,extraction ratio of material to liquid of 1∶45,extraction time of 3.5 h.The extraction rate of content polysaccharide was 3.99%.The monosaccharide composition of polysaccharide from Phellinus igniarius was initially identified as D-glucose,D-galactose,Arabia L-sugar and D-lactose.
