摘要
以耐寒柑橘(Critus reticulata Blanco)为试材,研究了培养基、消毒处理方式、外植体类型以及2,4-D与KT对柑橘愈伤组织诱导的影响,优化柑橘愈伤组织培养条件.结果表明,柑橘愈伤组织诱导中MT培养基的诱导效果优于MS培养基,统计学分析差异显著;带外种皮消毒处理方式更有利于外植体的脱分化,愈伤诱导率均在90%以上;胚与子叶外植体分别在f与i培养基上脱分化效果相对最佳,其诱导率为93.10%与93.33%;培养基中2,4-D浓度为2.0-3.0 mg/L适合柑橘愈伤组织的诱导,且激素KT与2,4-D之间具有良好的正向协同作用,当培养基中KT为0.5 mg/L时,诱导率增加25.75%,疏松愈伤率增加14.48%.
In order to optimize the culture conditon of cold-resistant Critus reticulata Blanco,the effects of different factors including medium,sterilize ways,explants types,2,4-D and KT exerted on callus induction were studied.The results showed that inducing rate of MT was better than that of MS,proving significant influence by independent-sample.t test.Way of sterilizing with testa was an appropriate pretreatment because it was conductive to callus forming with induction rate of more than 90%.Optimal medium for embryo and cotyledon were f and i with induction rate of 93.10% and 93.33%,respectively.The suitable mass concentration for 2,4-D was 2.0-3.0 mg/L.There was positive synergistic effect between 2,4-D and KT.Induction rate and loose callus rate increased 25.75%,14.48% when KT was 0.5 mg/L in the medium.
