Simultaneous Determination of Scutellarin,Protopine,Apigenin and Tetrahydropalmatine in Dengyan Granules by HPLC

摘要 目的:建立HPLC同时测定灯延颗粒中灯盏乙素、原阿片碱、芹菜素和延胡索乙素含量的方法。方法:采用Zorbax Extend-C18色谱柱(4.6 mm×250 mm,5μm),流动相乙腈(A)-0.3%醋酸铵水溶液(B),梯度洗脱(0~25 min,15%A;25~70 min,15%~65%A;70~75 min,65%~15%A),流速0.8 m L·min^-1,柱温30℃,检测波长分别为280 nm(灯盏乙素、原阿片碱、延胡索乙素)、337 nm(芹菜素)。结果:灯盏乙素、原阿片碱、芹菜素和延胡索乙素进样量分别在0.90~8.99,0.73~7.35,0.05~0.54,0.14~1.37μg线性关系良好;平均回收率分别为99.48%,98.17%,97.41%,98.33%,RSD分别为0.8%,1.2%,1.2%,0.5%。结论:该法适用于同时测定灯延颗粒中灯盏乙素、原阿片碱、芹菜素和延胡索乙素的含量。 Objective: The aim of this study was to develop an HPLC method for the simultaneous determination of contents of scutellarin, protopine, apigenin and tetrahydropalmatine in Dengyan Granules. Method: The analysis was performed on a Zorbax Extend-C18 column (4. 6 mm× 250 ram, 5 μm) in gradient elution (0-25 rain, 15%A; 25-70 rain, 15%-65% A; 70-75 rain, 65%-25% A) by using the mobile phase of acetonitrile (A) -0.3% ammonium acetate solution (B). The flow rate was 0. 8 mL ·min^-1 and the column temperature was maintained at 30 ℃ , the detection wavelengths were set at 280 nm (for scutellarin, protopine and tetrahydropalmatine) , 337 nm (for apigenin) , respectively. Result : The calibration curves were linear within the range of 0.90-8.99 Ixg for scutellarin, 0.73-7.35 ixg for protopine, 0.05-0.54 p,g for apigenin and 0. 14-1.37 p,g for tetrahydropalmatine, respectively. The recoveries of scutellarin, protopine, apigenin and tetrahydropalmatine were 99.48% ( RSD 0. 8% ), 98. 17% ( RSD 1.2% ), 97.41% ( RSD 1.2% ) and 98.33% ( RSD 0.5% ), respectively. Conclusion: The method can be used for simultaneous determination of contents of scutellarin, protopine, apigenin and tetrahydropalmatine in Dengyan Granules.
