知母盐制前后降血糖作用及其机制 被引量:34

Study on hypoglycemic effects of Rhizoma Anemarrhenae before and after processed with saltwater and its mechanism

摘要 目的:比较知母盐制前后降血糖作用,并探讨其作用机制。方法:采用HPLC法,以对-硝基苯基-α-D-吡喃葡萄糖苷(PNPG)为底物,分析水解产物中对-硝基酚(PNP)的浓度,比较生知母和盐知母对α-葡萄糖苷酶的抑制作用;采用STZ诱导2型糖尿病大鼠模型,以阿卡波糖为阳性对照,比较生知母、盐知母水煎液对糖尿病大鼠空腹血糖(FBG)水平的影响,进行口服葡萄糖耐量试验,并采用ELISA法检测糖化血红蛋白(HbA1c)和血清胰岛素(FINS)水平,并计算胰岛素抵抗(Homa-IR)指数和胰岛素敏感(ISI)指数。结果:生知母和盐知母水煎液对α-葡萄糖苷酶有较强的抑制作用,呈浓度正相关,且盐知母优于生知母;各给药组均能明显降低FBG、HbA1c、FINS、Homa-IR,升高ISI,改善糖耐量。其中除对ISI作用外,盐知母均优于生知母,且盐知母对FINS的作用强于阳性对照药阿卡波糖。结论:生知母、盐知母均能抑制α-葡萄糖苷酶活性,对2型糖尿病大鼠糖代谢有改善治疗作用,盐制后作用显著增强。 OBJECTIVE To compare the hypoglycemic effects of Anemarrhenae Rhizoma before and after processed with salt-water and explore its mechanism. METHODS P-nitrophenyl-α -D-glucopyranoside(PNPG) was used as substrate and the P-nitrophenol(PNP) concentration was analyzed by HPLC to compare the a glucosidase inhibition effect between Anemarrhenae Rhizorna and salt Anemarrhenae Rhizoma ; the rats induced by STZ were epmployed as diabetes control group while those trea- ted with acarbose as the positive group. The effects of Anemarrhenae Rhizoma and salt Anernarrhenae Rhizoma decotion on fasting blood glucose(Fl3G) and oral glucose tolerance were evaluated. The glycosylated hemoglobin(HbAlc) and fastings erum insulin(FINS) were measured by ELISA. The index of insulin resistance (Homa-IR) and insulin sensitivity index (ISI) were ca- culated. RESULTS Anemarrhenae Rhizoma and salt Anemarrhenae Rhizoma decotion have a strong inhibitory effect on a-glu- cosidase,which was positively correlated with the concentration, the effect of Anemarrhenae Rhizoma was stronger than Anenu^rrhenae Rhizoma. Each treatment group can significantly reduce the FBG, HbAlc, FINS, Homa-IR, increase ISI and im- prove glucose tolerance. The effect of salt Anemarrhenae Rhizoma on FINS was stronger than positive control group. CON- CLUSION Anernarrhenae Rhizorna and salt Anemarrhenae Rhizoma both can inhibit ,- glucosidaso activity and iranrnvo thor- apeutic effect on glucose metabolism in type Ⅱ diabetic rats. They both can significantly enhance hypoglycemic effect after pro- cessing.
