摘要
目的:探讨四物汤有效成分与脂质体模拟生物膜的相互作用,以了解其有效成分体内吸收的情况。方法:采用平衡透析与高效液相色谱-质谱联用技术,对四物汤有效成分与脂质体模拟生物膜相互作用进行分析;同时考察了透析时间、脂质体溶液的浓度和缓冲溶液p H对四物汤有效成分与模拟生物膜相互作用的影响。结果:四物汤中有9个成分与脂质体相互作用明显,通过对照品对照及LC-MS方法鉴定了其中6个成分,分别是香草酸、芍药内酯苷、毛蕊花糖苷、芍药苷、洋川芎内酯Ⅰ、肉桂酸;模拟生物膜的浓度对四物汤有效成分与模拟生物膜相互作用影响较大,随着浓度增加,色谱峰面积明显减小,符合被动吸收规律;p H对其中4个成分有明显影响。结论:脂质体平衡透析液质联用色谱法可作为快速筛选中药潜在的活性成分的有效方法,为药效物质基础的进一步研究提供参考。
Objective:To explore the interaction between the effective components of Siwu decoction and liposome biomembrane,in order to understand in vivo absorption of the effective components. Methods:The interaction of the effective components of Siwu decoction with liposome biomembrane was analyzed by the equilibrium dialysis and LC - MS coupling technique. The effect of experiment conditions on the interaction was investigated, including the dialysis time, the liposome concentration, and the pH value of buffer. Results:Nine compounds in Siwu decoction had obvious interaction with liposome. Among them, six compounds were identified by comparison with the standards and LC - MS method, i. e. , vanillic acid, albiflorin, acteoside, paeoniflorin, senkyunolide I and cinnamic acid. The concen- tration of liposome remarkably affected the absorption of most compounds as the peak area was significantly de- creased along with the increase of liposome concentration, which was in accordance with passive absorption pattern. The pH value had significant influence on four compounds. Conclusion:The established method contributes to the fast screening of the active components in traditional Chinese medicine and providedes reference for further research of pharmacodynamic material basis.
