复脉膏外敷对大鼠长春瑞滨外渗性损伤皮肤中EGFR的影响 被引量:2

Effects of Fumai Paste on EGFR of Skin in Vinorelbine-induced Extravasation Injury Model Rats

摘要 目的:建立大鼠长春瑞滨外渗性皮肤损伤的理想实验模型,在此动物模型上研究复脉膏外敷对外渗性损伤皮肤中EGFR的影响,以探讨复脉膏外敷治疗化疗性静脉炎的疗效及机制。方法:42只大鼠随机分为两组:复脉膏组、50%硫酸镁组,21只/组,建立实验模型。两组大鼠左侧损伤处分别予复脉膏外敷、50%硫酸镁湿敷;右侧予生理盐水外敷作为对照组。在损伤后第1、4、7、11、14、18天,分别随机取3只大鼠,切除损伤部位的皮肤及皮下组织,固定标本,脱水,石蜡包埋,连续切片,采用免疫组化标记及图像分析方法,在电子显微镜下观察EGFR的表达情况。结果:EGFR阳性表达主要见于表皮细胞、真皮成纤维细胞、毛囊上皮细胞、血管内皮细胞、巨噬细胞、中性粒细胞的胞膜及胞浆中。复脉膏外敷较硫酸镁湿敷及生理盐水外涂能更早更强刺激长春瑞滨外渗性损伤皮肤中EGFR的阳性表达(P<0.05)。结论:复脉膏外敷于损伤皮肤后能显著促进EGFR的快速增加来促进肉芽组织及鳞状上皮的生成,进而促进损伤皮肤的愈合。因此,复脉膏可以用于防治化疗性静脉炎。 Objective: To establish an ideal animal experiment model of vinorelbine-induced extravasation injury to study on effects of Fumai Paste on EGFR of skin, then discussing the curative effect and mechanism of treatment of chemotherapeutic phlebitis with external application of Fumai Paste. Methods: Forty-two rats were divided randomly into two groups with 21rats in each group. Each rat was established experimental model. Each rat of group of Fumai Paste was treated with external application of Fumai Paste on left side; each rat of group of magnesium sulfate was treated with hydropathic compress of 50% magnesium sulfate on left side. The right side of each rat were controlled group, and were treated with hydropathic compress of normal saline. Three rats were selected randomly from each group in lday, 4days, 7days, lldays, 14days and 18days, and the skin and subcutaneous tissue of extravasation injury of each rat was cut off. All specimens were fixed in formalin, were dehydrated, embedded in paraffin and serially cut. All specimens were observed expressions of EGFR under the electron microscope by immunohistochemical assay and image analysis method. Results : The positive expression of EGFR was major in epidermic cells, dermal fibroblast, hair follicle cells, vascular endothelial cell, macrophage, cell membrane and endochylema of nentrophile granulocyte. Fumai Paste could stimulate the positive expression of EGFR of skin of animal experiment model of vinorelbine-induced extravasation injury more earlier and more strong than 50% magnesium sulfate and normal saline (P〈0. 05). Conclusion : Fumai Paste can remarkable promote the increase of EGFR of skin of animal experiment model of vinorelbine-induced extravasation injury. Then with the generation of granulation tissue and squamous epithelium, Fumai Paste can accelerate the healing of the damaged skin. So chemotherapeutic phlebitis can be prevented and treated by external application of Fumai Paste.
