四神丸改善TNBS及DSS诱导小鼠实验性结肠炎的研究 被引量:12

摘要 目的观察四神丸对三硝基苯磺酸(TNBS)和低分子量硫酸葡聚糖钠(DSS)诱导的小鼠实验性结肠炎的防治作用。方法小鼠麻醉后直肠内灌注含1.5 mg TNBS的50%乙醇溶液制备TNBS结肠炎模型;小鼠自由饮用含3%DSS的蒸馏水5 d制备DSS结肠炎模型。各模型均在造模第2天开始灌胃给四神丸,连续8 d。观察小鼠的一般状态和结肠炎疾病活动度指数(DAI)评分、结肠组织学及结肠中髓过氧物酶(MPO)变化。结果 TNBS及DSS模型小鼠DAI评分及结肠组织中MPO活性显著高于对照组,模型小鼠可见结肠黏膜上皮脱落、大量炎细胞浸润及腺管扭曲等。经四神丸治疗后能显著改善TNBS及DSS结肠炎小鼠的一般状况及DAI评分,并能改善结肠的组织学损伤、减轻炎细胞的浸润。结论四神丸对TNBS及DSS所诱导的小鼠结肠炎均有显著的改善作用。 Objective To explore the effects of Sishen pill on colitis model induced by trinitrobenzene sulfonic acid (TNBS) or dextran sulfate sodium (DSS) in mice. Methods TNBS colitis model (TNBS colitis) was induced by intrarectally administration of 1.5 mg TNBS in 50% ethanol. DSS colitis model (DSS colitis)was induced by feeding with 4% (w/v) DSS dissolved in drinking water. One day after modeling, mice were intragastically administrated with Sishen pill (2.3 g/kg) for 8 days. Changes in disease activity index (DAI) together with histological scores were evaluated, and myeloperoxidase (MPO) in colonic tissue was detected. Results Sishen pill significantly reduced DAI and histological scores of mice in both TNBS colitis and DSS colitis, it also alleviated inflammatory cell infiltration of colonic tissue in both models. Conclusion Amelioration efficacy of Sishen pill on mice with TNBS colitis and DSS colitis were obtained.
