摘要 目的:克隆一个水稻β-肌动蛋白基因,并研究其蛋白的亚细胞分布。方法:利用RT-PCR技术从日本晴水稻中扩增得到β-肌动蛋白基因Os Actin,将其与拟南芥肌动蛋白基因家族进行分子进化分析;利用胶体金免疫电镜技术对水稻β-肌动蛋白基因进行定位分析。结果:核苷酸和氨基酸序列分析表明,Os Actin与水稻的Os RAc1基因有较高的同源性(98%);胶体金免疫电镜定位结果显示β-肌动蛋白在细胞核、细胞质中均有分布。结论:获得了水稻β-肌动蛋白基因Os Actin,并研究了其蛋白在水稻韧皮部内的分布,为进一步探讨该基因的功能及作用机理奠定了基础。 Objetive: To clone a rice β-actin gene and study the subcellular distribution of its protein. Meth?ods: A β-actin gene OsActin was amplified by RT-PCR from Nipponbare rice plant. Its molecular evolution was analyzed by comparing with the β-actin gene family from Arabidopsis thaliana and then its subcellular localization in rice was determined by colloidal gold immunoelectron microscopy technique. Results: Sequence analysis re?vealed that OsActin shared high homology(98%) with OsRAc1 gene at the nucleotide and amino acid levels. The results of immunogold electron microscopy showed that β-actin was distributed in both nucleus and cytoplasms of rice plant. Conclusion: The OsActin gene was obtained and the distribution of β-actin was studied in rice phlo?em. These results laid a foundation for further study on the function and mechanism of OsActin.
