摘要
利用气相色谱方法测定挥发性脂肪酸,运用单因素试验与正交试验相结合,对气相色谱测定挥发性脂肪酸的条件进行了初步优化。结果表明:使用FFAP毛细管色谱柱(30 m×0.32 mm×0.5μm)测定挥发性脂肪酸的最佳条件为:进样量1.5μL,载气流量75 m L/min,柱温条件:程序升温(初始温度100℃、停留2 min,以6℃/min升至130℃、停留1 min,再以10℃/min升至190℃、停留2 min),进样口温度220℃,检测器温度230℃。在该条件下,测定的挥发酸组分可以完全分离且分析速度快,峰型基本对称。
A gas chromatographic method was used to determine volatile fatty acid, and the determination condition was optimized by orthogonal test in combination with single factor test. The results showed that the optimum detection parameters were as follow: sample volume of 1.5 μL, carrier gas flow rate of 75 mL/min, injection and detector temperatures at 220℃, 230℃ respectively, the column temperature conditions: temperature programmed (initial temperature of 100℃ for 2 min, with CC/min to 130% for 1 rain, with 10℃/min to 190℃ for 2 min), by using FFAP capillary-column chromatography (30 m×0.32 mm× 0.5 μm). Under these conditions, the volatile acid component could be completely separated in quickly and substantially symmetrical peak way.
