上皮间质转化在食管鳞状细胞癌转移中的作用 被引量:1

摘要 目的观察上皮间质转化相关标记物E-钙黏蛋白(E-cadherin)和Vimentin基因及其蛋白在食管鳞状细胞癌淋巴结转移组和非转移组中的表达,并分析E-cadherin和Vimentin表达的相关性。方法采用免疫组织化学SP法和实时定量逆转录聚合酶链反应(qRT-PCR)检测47例有淋巴结转移、30例无淋巴结转移的食管癌和20例正常食管组织中E-cadherin和Vimentin基因及其蛋白的表达情况。结果对照组、非转移组与转移组Ecadherin蛋白表达分别为20/20(100%)、29/30(96.67%)和44/47(93.62%),组间比较差别有统计学意义(P<0.05)。转移组的高表达率22/47(46.8%)低于非转移组18/30(60%),但差别无统计学意义(P>0.05)。Vimentin蛋白在对照组、非转移组、转移组阳性表达率逐渐增高,分别为0/20(0)、11/30(36.67%)、37/47(78.72%),组间比较差别有统计学意义(P<0.05),转移组与非转移组间比较差别有统计学意义(P<0.05)。Vimentin和E-cadherin表达呈显著负相关,相关系数(r=-0.377)。qRT-PCR检测显示非转移组和转移组中E-cadherin和Vimentin mRNA表达量与对照组比较差别有统计学意义(P<0.05)。E-cadherin基因的表达在转移组(0.43±0.23)与非转移组(0.51±0.54)间的差别无统计学意义(P>0.05),而转移组Vimentin表达(4.05±1.88)明显高于非转移组(2.70±1.73),差别有统计学意义(P<0.05)。结论上皮间质转化相关基因E-cadherin和Vimentin与食管鳞癌的转移密切相关,可作为预测食管癌转移复发及评估临床预后的有效标记物。 Objective To identify the differences of epithelial mesenchymal transition (EM T ) re-lated biomarkers of E-cadherin and Vimentin in mRNA and protein expression levels between esophageal squamous cell carcinoma (ESCC) with lymph node metastasis and no lymph node metastasis . Methods Immunohistochemistry SP method and quantitative reverse transcriptase real-time polymerase chain reac-tion (qRT-PCR) array were applied to detect mRNA and protein expression levels of E-cadherin and Vim-entin on 47 cases of ESCC with lymph node metastasis and 30 cases of ESCC without lymph node metasta-sis ,and 20 cases of normal esophageal mucous membrane tissues . Results The percentage of expressing E-cadherin in normal control group ,non metastasis and metastasis group of ESCC were 20/20 (100% ) , 29/30 (96 .67% ) ,and 44/47 (93 .62% ) respectively ,there was a statistically significant difference among three groups ( P〈0 .05) ,but the percentage of high E-cadherin expression of metastasis group (22/47 , 46 .8% ) and non metastasis group (18/30 ,60% ) had no significant difference (P〉0 .05) . The number of cases with the positive Vimentin protein expression increased in order of normal control group ,no me-tastasis group and metastasis group ,the percentage was (0/20 ,0) ,(11/30 ,36 .67% ) ,(37/47 ,78 .72% ) respectively ,showing a significant difference between two groups (P〈0 .05) . The rate with high ex-pression level of Vimentin between the two groups of metastasis group (11/47 ,23 .4% ) and no metastasis (2/19 ,10 .53% ) had statistical significance (P〈0 .05) . There was a significant negative correlation be-tween Vimentin and E-cadherin ,the correlation coefficient was r= -0 .377 . qRT-PCR analysis showed that there was a obvious difference in mRNA expression levels of E-cadherin and Vimentin in the normal group ,no metastasis group and metastasis group of ESCC (P〈 0 .05) . E-cadherin mRNA expression level showed no significant difference between metastasis group (0 .43 ± 0 .23) and non metastasis group (0 .51 ± 0 .54)(P〉 0 .05) ,while that of Vimentin had a significant difference between metastasis group (4 .05 ± 1 .88) and non metastasis group (2 .70 ± 1 .73) (P〈 0 .05) . Conclusions The expressions of EM T related genes E-cadherin and Vimentin were closely correlated with metastasis of ESCC ,and there-fore could be used for predictors of the metastasis and recurrence of ESCC as well as useful biomarkers to prognosis .
