大黄虫丸对Lewis肺癌小鼠血管生成的影响及机制 被引量:15

摘要 目的:通过观察大黄虫丸对Lewis肺癌小鼠血管内皮生长因子(VEGF)、环氧化酶-2(COX-2)表达的影响,及其对微血管生成的作用,探索其抗肿瘤的可能机制。方法:造模后次日先随机分成5组,每组10只。根据预试结果确定高(H)、中(M)、低(S)剂量;同时设空白组和化疗组。d11处死小鼠,剖取完整肿块称重并计算抑瘤率。以免疫组化法检测VEGF、COX-2的表达,并用血管内皮细胞标记物CD34单克隆抗体标记行免疫组化染色,观察肿瘤微血管生成状况。结果:实验组高(H)、中(M)、低(L)和化疗组抑瘤率分别为29.70%、38.79%、17.58%、63.64%。免疫组化法表明大黄虫丸具有抑制VEGF、COX-2表达的作用,同时降低肿瘤微血管生成。结论:大黄虫丸对荷Lewis肺癌小鼠肿瘤具有明显的抑制作用,作用机制可能与抑制VEGF及COX-2表达,降低肿瘤微血管生成有关。 Objective:By observing effects of Dahuang Zhechong Pills on VEGF, Cox -2 and the tumor micrangium inmice with Lewis Lung carcinoma to discuss its mechanism of anti - tumor. Methods : The mice were divided randomly intofive groups the day after modeling and each group included ten mice. According to the pre -test, high -dose, medium -dose and low- dose were decided, at the same time the model group and chemotherapy group were set up. All mice werekilled,weighted and compared the tumor and we calculated tumor growth inhibition rates of each group on the 12th day.hnmunohistochemical staining was use to check the protein expressions of VEGF and COX - 2 and the tumor micrangiumwas also examined by using CD34 staining. Results:The inhibition rates in experimental groups of high (H), medium (M)and low (L) dosages and the chemotherapy group were respectively 29.70% ,38.79% , 17.58% and 63.64%. The re-sults fi'om immunohistochemical staining showed that the protein expressions of EGFR and COX - 2 were reduced and thetumor micrangium was also dropped. Conclusion:Dahuang Zhechong Pills inhibited Lewis lung carcinoma in C57BL mice,which may depend on reducing the protein expressions of EGFR and COX - 2 and reducing the tumor angiogenesis.
