摘要
目的:探讨Heparanase、COX-2在肾癌中表达的相关性及与肿瘤血管生成的关系及意义。方法:采用免疫组化PV6000法检测54例肾癌和20例正常肾脏组织中Hpa、COX-2蛋白的表达,以CD34单克隆抗体进行微血管内皮染色,计算微血管密度,分析Hpa、COX-2在肾癌中表达的相关性以及与临床病理特征及微血管密度的关系。结果:Hpa、COX-2在肾癌中的表达均较正常肾脏组织明显增高(P<0.05)。Hpa的表达与肾癌的临床分期、肿瘤大小、远处转移有关(P<0.05),而与患者年龄、性别、肿瘤的分化程度无相关性(P>0.05)。COX-2的表达与肾癌的临床分期、远处转移及分化程度有关(P<0.05),与患者年龄、性别、肿瘤大小无关(P>0.05)。肾癌组织中MVD显著高于正常肾脏组织(P<0.05),高MVD组中Hpa、COX-2表达的阳性率高于低MVD组(P<0.05),Hpa、COX-2阳性表达组中MVD值亦高于阴性表达组(P<0.05)。Hpa、COX-2的表达具有相关性(P<0.05)。结论:Hpa、COX-2可能通过促肿瘤血管生成参与肾癌的恶性生物学行为,并有可能成为判断肾癌预后的指标和靶向治疗的靶点。
Objective:To investigate the expression parterns of Heparanase and COX-2 protein in renal cell car-cinoma and their correlations with tumor progression and angiogenesis. Methods:The expressions of Heparanase and COX-2 in 54 renal cell carcinoma and 20 normal renal tissues were assessed by immunohistochemistry and the corre-lation with clinicopathologic features and microvessel density. Results:The expression of Heparanase and COX -2 protein significantly increased in renal cell carcinoma compared with that in normal renal tissues(p〈0. 05). The ex-pression of Heparanase protein was positively correlated to the clinical stages,tumor diameter and distant metastasis of RCC(p〈0. 05). There were no associations between the expression of Heparanase and age,gender and differentiation (p〉0. 05). The expression of COX-2 protein was positively correlated to the clinical stage,differentiation and dis-tant metastasis of RCC(p〈0. 05). There were no associations between the expression of COX-2 protein and tumor diameter,age and gender. RCC specimens had higher MVD than normal renal tissues(p〈0. 05). Patients who showed high MVD was significantly associated with Heparanase and COX-2 positive tumors(p〈0. 05). Heparanase and COX-2 positive tumors had a significantly higher MVD compared with negative tumors(p〈0. 05). Expression of Heparanase was significantly correlated with that of COX-2(p〈0. 05). Conclusion:Heparanase and COX-2 may be involved in malignancy of RCC through promoting tumor angiogenesis. It may be prognostic factors and therapeutic target of RCC.
