靶蛋白Xklp2和胸苷磷酸化酶在妇科肿瘤中的研究进展

摘要 恶性肿瘤是严重危害人类健康的疾病,2007年全球有790万人死于癌症,占全球人类死亡总数的13%,居全球死亡原因第1位[1].卵巢癌、宫颈癌、子宫内膜癌为妇科3大恶性肿瘤,其发病率近年来在世界范围内呈上升趋势,严重威胁广大妇女的生命健康.肿瘤的发生发展是多因素共同作用的结果,其机制复杂,可能与癌基因活化、抗凋亡作用及新生血管形成等多种因素有关.靶蛋白Xklp2(TPX2)与胸苷磷酸化酶(TP)作为新近发现的癌基因日渐显示出其对恶性肿瘤机制研究的优越性.二者的发现改变了人们对DNA主导生命现象的观念.TPX2也称REPP86,发现于蟾蜍卵细胞,是一种受细胞周期严格调控的微管相关蛋白,弥漫分布在细胞周期的S期和G2期,在有丝分裂的纺锤体形成过程中发挥重要作用,是一种新的癌基因[2],其过度表达可导致中心体扩增,出现DNA异倍体及多倍体,促进肿瘤细胞的分裂和血管生成,与细胞癌变和恶性肿瘤的进展有关.
