生长抑素受体亚型mRNA在胃癌组织中的表达

摘要 目的检测生长抑素受体(Somatostatin receptor, SSTR)各亚型在人胃癌组织及其癌旁组织中分布和表达,并探讨其表达与胃癌的临床及病理学特点之间的关系及其意义.方法:随机收取经病理确诊的50 例人胃癌组织及其癌旁组织,并收集其相关临床病理资料,采用RT-PCR 法检测SSTR1~5 各 mRNA 亚型在胃癌组织及其癌旁组织中表达,采用捷达810 分析软件对结果进行相对定量分析.结果:50 例胃癌患者组织中均有SSTR 基因的表达,SSTR1、SSTR2、SSTR3、SSTR4 和SSTR5mRNA 亚型表达阳性率在胃癌组织中分别为:32%(16/50)、100%(50/50)、70%(35/50)、68%(34/50)和68%(34/50);在癌旁组织中分别为:40%(20/50)、 92%(46/50)、 84%(42/50) 、46%(23/50)、 38%(19/50);SSTR4 和SSTR5 mRNA 亚型在胃癌组织中的表达高于癌旁组织的表达(P〈0.05);采用相对定量分析, SSTR2、SSTR4 和SSTR5 mRNA 亚型在胃癌组织中的表达高于癌旁组织中的表达(P〈0.05).SSTR3 mRNA 在高/中分化胃癌组织中的表达高于低分化胃癌组织中的表达(P〈0.05).结论:胃癌组织中均有一种以上生长抑素受体亚型基因的表达,其中以SSTR2 mRNA 表达率为最高,为临床选择应用SSTR 高特异性和高亲和性SSTA 治疗胃癌提供依据.SSTR3 mRNA 的表达与临床病理特征有关,其可能在胃癌的发生、发展过程中发挥一定的作用. ObjectsTo investigate SSTR subtypes mRNA expression in tissue of gastric cancer ,and explore its relationship with clinicopathological characters. Methods:RT-PCR was performed to detect the expression of SSTR 1~5 subtype mRNA in tissues of gastric cancer and adjacent gastric mucosa of 50 cases collected, and including relatedclinical data. The relative quantification was analyzed by using Jieda 801 analysis software.Results: there were SSTR mRNA expression in 50 tissues of gastric cancer. Positiveexpression rate SSTR1、SSTR2、SSTR3、SSTR4 and SSTR5 mRNA in tissues of gastric cancer were 32%(16/50)、100%(50/50)、70%(35/50)、68%(34/50) and 68%(34/50) respectively,and in tissues of adjacent gastric cancer were 40%(20/50)、 92%(46/50)、 84%(42/50) 、46%(23/50) and 38%(19/50) respectively. Expression rate of SSTR4 and SSTR5 mRNAsubtypes in tissues of gastric cancer was higher than in tissues of adjacent cancer(P〈0.05). Expression rate of SSTR2、SSTR4 and SSTR5 mRNA subtypes in tissues of gastric cancerwas higher than in tissues of adjacent cancer by the relative quantification. Expression of SSTR3 mRNA in high/middle differentiated tissues was lower than poorly differentiatedtissues of gastric carcinoma. Conclusion:There is more than one of SSTR subtypes mRNA expression in tissue of gastric carcinoma. Expression of SSTR2 mRNA is the highest. Theresults provide a basis for clinical choicing SSTA to treat gastric carcinoma with SSTR high affinity and specificity. Expression of SSTR3 mRNA is relationship with clinicopathologicfeatures. It play a important role in carcinogenesis and progression of gastric cancer.
