缺氧预处理对大鼠皮层神经元缺血耐受性和NF-κB表达的影响

Effects of ischemic-preconditioning on ischemic tolerance and NF-κB expression in cultured rat cortical neurons

摘要 目的观察缺氧预处理对大鼠原代神经元缺氧耐受性的影响,并探讨缺氧预处理对核转录因子κB(nuclear factor -κB,NF-κB)表达的影响.方法:体外培养的原代神经元分为3 组:正常对照组、再灌注组和预处理组,再灌注组给予类缺血30min 复氧培养24h;预处理组经类缺血10min 预处理后复氧培养24h 后,再给予致死性的类缺血30min 复氧培养24h,三组细胞均用台盼蓝染色,检测神经元坏死,同时用免疫组化法观察三组细胞NF-κB 的表达.结果:再灌注组神经元的存活率明显低于预处理组,再灌注组NF-κB 的表达明显低于预处理组,差异有显著性;正常对照组与预处理组差异无显著性.结论:缺氧预处理能够增加神经元的缺氧耐受性,预处理可能通过激活NF-κB 发挥神经保护作用. Objective To study the effects of ischemic preconditioning on ischemic tolerance and the expression of NF-κB in primary cultured rat cortical neurons. Methods:The neurons primary cultured in vivo were randomly divided into 3 groups: control group (CG)、 ischemia-reperfusion(I/R) and ischemic preconditioning group (IP). I/R were treatedwith 30min ischemia and IP were treated with 10min ischemic preconditioning before 30 min ischemia. The necrosis of two group cells was detected by trypan blue. NF-κB expressionswere studied with immunohistochemical assay. Results: The survival rate in I/R was lower than it in IP. The expression of NF-κB in I/R was obviously lower than it in IP. And there wassignificant difference between I/R and IP. Conclusion: Ischemic preconditioning can induce ischemic tolerance in cultured cortical neurons. Preconditioning might activate NF-κB andadd to its expression to play neuroprotection.
