基因沉默HSP90基因促进肺癌耐药细胞株凋亡作用研究

摘要 目的通过基因沉默HSP90 基因后观察肺癌耐药细胞的耐药逆转情况;方法：siRNA 转染肺癌耐药细胞A549/DDP,实时荧光定量PCR 和western 分别检测HSP90 在基因水平和蛋白水平表达的改变,再进一步检测细胞活性,凋亡以及caspas 3 的改变;结果：siRNA 转染成功下调细胞中HSP90 基因在基因和蛋白水平的表达,表达下调后,耐药细胞对顺铂的敏感度增加,细胞活性下降,凋亡增加;结论：通过基因沉默HSP90 基因能够有效的逆转A549/DDP 细胞对顺铂的耐药性.
